To this end, the ability to directly measure the immune response (in particular, T-cell mediated) in a sample of interest is an important technique for monitoring an individual's immune response. However, this goal has proven to be extremely difficult, particularly in the case of T-cell mediated immune responses. In response to disease, the immune system is generally capable of mounting an initial response, or primary response, to a disease-related antigen. This initial response is generally robust and protective. However, for most individuals the immune system fails to maintain an effective long-term immune response. Thus, most diseases recur and most individuals, once infected, will suffer from repeated or persistent infections. One reason for the inability of the immune system to maintain a functional long-term immune response may be that the required T cells are not generated in sufficient numbers. To date, the most reliable method of measuring the effectiveness of a T-cell mediated immune response has been to measure the level of antigen-specific antibodies secreted by the B cells in the individual. This is because the presence of antigen-specific antibodies in an individual usually indicates that at least one T cell has been activated by the B cell. More specifically, the presence of an antigen-specific antibody in a given sample indicates that the B cell successfully activated a T cell. While the presence of antigen-specific antibodies, particularly antigen-specific IgG, can indicate the presence of activated T cells, it cannot indicate if the activated T cells are of a type that are required for maintaining the long-term immune response. Furthermore, the presence of an antigen-specific antibody alone can be insufficient to indicate whether the T-cell mediated immune response is functional. In some cases, the B cells may not successfully activate the required T cells, or the activated T cells may not be required to maintain the immune response. There is a need in the art for improved techniques for measuring the effectiveness of T-cell mediated immune responses. The present invention addresses this
